Establishment of hybridoma cell lines secreting anti-Flavobacterium columnare monoclonal antibody and identification of their immunological properties

Abstract
To prepare for monoclonal antibody(McAb) against Flavobacterium columnare,BALB/c mice were immunized with a particle antigen.The spleen cells from the immunized mice were fused with mouse myeloma cells SP2/0.Two hybridoma cell lines(2B5 and 4G7) stably secreting McAb against F.columnare were obtained by limited dilutions.The antibody secreted belonged to IgG1 subtype with a κ light chain.No cross-reactions were observed between the McAb prepared and other aquatic pathogenic bacteria such as Edwardsiella tarda,Escherichia coli,Aeromonas hydrophila,Vibrio anguillarum,Vibrio alginolyticus,Vibrio parahaemolyticus and Vibrio harveyi.The method was established successfully in generating McAb against F.columnare for the first time,which provides the foundation for the rapid detection of F.columnare.关键词
