Abstract
Objective To investigate the protective effect of isoflurane on the injury of human umbilical vein endothelial cells induced by hydrogen peroxide in vitro and to explore its underlying mechanism.Methods Human umbilical vein endothelial cells were cultured in vitro and were divided into three groups:normal control group (group A),isoflurane group (group B),isoflurane and hydrogen peroxide group (group C) and hydrogen peroxide injury group (group D).Flow cytometry was used to detect cell apoptosis.Western blot analysis was performed to detect HSP27 protein expression.Reverse transcriptase PCR was used to detect HSP27,Bcl-2 and Caspase-3 mRNA expression in the endothelia cells.Results Compared with normal control group,hydrogen peroxide cause endothelial cell apoptosis (P < 0.05).Isoflurane can reduce endothelial cell apoptosis induced by hydrogen peroxide (P < 0.05).HSP27 and Bcl-2 expression increased,while Caspase-3 decreased in isoflurane group.Conclusions Isoflurane can inhibit human umbilical vein endothelial cells apoptosis induced by hydrogen peroxide,the mechanisms may be associated with increased expression of HSP27 protein and Bcl-2 and with decreased expression of caspase-3.关键词
