Effects of EGCG on the malignant behavior and the expression of MT1-MMP of breast cancer cell line MCF-7

Abstract
Objective To investigate the effects of epigallocatechin gallate (EGCG) on the malignant behavior of breast cancer cell and the possible mechanisms.Methods MCF-7 cell line transfected by the MT1-MMP eukaryotic expression vector was established first.Then the expression of MT1-MMP protein,the breast cancer cell line invasion ability,and the cell proliferation ability was respectively detected by Western blot,Transwell method,and cell counting kit-8 (CCK-8).The MCF-7 cells were divided into 3 groups:non-transfected group,null vector transfection group,and transfection group.The results of all the groups were compared and analyzed.Results The expression of MT1-MMP protein was inhibited after EGCG transfection.The invasion and proliferation ability of the MCF-7 cell line expressing MT1-MMP protein were obviously higher than those of the non-transfected group and null vector transfection group,which were inhibited after EGCG transfection in a concentration-dependent manner.Conclusion EGCG had an inhibiting effect on the invasion and proliferation ability of breast cancer cells and the mechanism was probably related to the inhibition of the expression of MT1-MMP protein.关键词
