GM130, a cis-Golgi protein, regulates spindle assembly during mouse oocyte meiotic maturation

Abstract
Objective:To identify the role of GM130,a cis-Golgi protein,in regulating meiotic spindle assembly.Methods:GM130 localization was determined by immunofluorescence and confocal microscopy.The expression of GM130 was detected by immunoblotting analysis.Results:The results showed that GM130 was localized to the spindle poles at both metaphase Ⅰ and metaphase Ⅱ stages and associated with the midbody at telophase Ⅰ stage.It showed that GM130 was expressed at all stages by immunoblotting.By nocodazole treatment,it clarified that GM130 localization was consistently dependent on spindle assembly.It also showed that knockdown of GM130 caused abnormal spindle formation and impaired the localization of microtubule organizing center (MTOCs) proteins γ-tubulin and Plk1.Conclusions:GM130 may act as a centrosome-associated protein to regulate spindle assembly.关键词
