骨桥蛋白在子宫内膜样腺癌中的表达及其与肿瘤增殖和患者预后的关系
Expression of osteopontin in endometrioid adenocarcinoma and its correlation with tumor proliferation and patient survival

Abstract
Objective To investigate the expression and clinical significance of osteopontin (OPN)in endometrioid adenocarcinoma.Methods Archived tissue samples of 30 cases of normal proliferative endometrium and 82 cases of endometrioid adenocarcinoma were selected and analyzed via IHC (immunohistochemically) with OPN as well as Ki-67,a main biologic index of tumor proliferation potentiality.Western blot was used to further validate the expression of OPN in carcinoma and adjacent tissues.Results IHC showed that OPN was overexpressed in endometrioid adenocarcinoma (63.4％),higher than that in normal proliferative endometrium,with statistical significance (P ＜ 0.05).Western blot also showedsignificantly higher OPN expression in endometrioid adenocarcinoma than in the matched adjacent tissue and normal endometrium.The expression of OPN was positively correlated with Ki-67 (r =0.393,P ＜0.01).With the increase of FIGO staging,histology grading,muscular invasion depth,the expression level of OPN gradually increased in endometrioid adenocarcinoma (all P ＜ 0.01).The higher expression of OPN was significantly associated with lower overall survival with logrank survival analysis (P ＜ 0.05).Conclusions OPN was overexpressed in endometrioid adenocarcinoma.It involved in tumor proliferation and is related with prognosis.It might be a new indicator of evaluating the malignancy and predict prognosis of endometroid carcinoma.关键词
