Abstract
Objective To observe vascular remodeling and angiogenesis and discuss the relationships between vascular remodeling with angiogenesis and the serum level of interleukin-8 (IL-8) in pulmonary hypertension (PH) rats.Methods Forty eight healthy male Wistar rats were randomly divided into control group (n =8) and monocrotaline (MCT)-treated group (n =40).The MCT group rats were intraperitoneal injected MCT 50 mg/Kg while the rats in control group were intraperitoneal injected an equal volume saline.The model group rats were drawn from day 7,4,21,28 (They were randomly divided into the 7,14,21,28 d groups with 10 Wistar rats of each group respectively).Control group was drawn from day 28.Rats in each group were measured right ventricular systolic pressure (RVSP)、right ventricular hypertrophy index (RVHI).The lung tissue was stained with hematoxylin and eosin to obtain the ratio of pulmonary small artery vessel wall area (vessel wall area%,WA%).The angiogenesis was investigated by CD105 immunohistochemistry of rat lung tissue to calculate the area ratio of the microvascular density (MVD).The serum IL-8 level was investigated by enzyme-linked immunosorbent assay (ELISA).Results Compared with the control group,RVSP RVHI and WA% of model group gradually increased with time extended.(7 d:P <0.05,14 to 28 d:P <0.01),MVD and the serum level of IL-8,showed an upward trend (7d:P < 0.05,14 to 28 d:P < 0.01),were higher than the blank control group at each time point (P < 0.01),especially at 14d.Correlation analysis showed that IL-8 and WA%,MVD,WA% and MVD had positive correlations (r =0.644,r =0.641,r =0.721,P < 0.01).Conclusion IL-8 involved in the pulmonary arterial remodeling and angiogenesis of the monocrotaline-induced PH in rats.The research explored new ideas for the mechanisms and treatment of PH.关键词
