Determination of 9 steroid hormones in human plasma and urine by HPLC

Abstract
AIM To develop a high performance liquid chromatography-ultraviolet (HPLC-UV)methed to determine 9 steroid hormones,including cortisol,cortisone,tetrahydrocortisone,tetrahydrocortisol,allo-tetrahydrocortisol,17β-estradiol,testosterone,progesterone and 17α-hydroxyprogesterone in human plasma and urine.METHODS Steroids in plasma were extracted and purified using SampliQ C1s ODS cartridge.Urine samples were processed by β-glucuronidase and collected by cartridge.The HPLC separation was carried out on an Ultimate* XB-C18(150 mm × 4.6 mm,5 μm) column with mobile phase of acetonitrile/water using a gradient elution mode at a flow rote of 1 mL· min-1.Column temperature was 35℃.Steroids were detected at wavelength of 205 nm and 245 nm.RESULTS The good linearity of 9 steroid hormones under chromatography was shown.Detection limits of 9 endogenous steroids ranged from 0.5-50μg·L-1.CONCLUSION HPLC-UV is sensitive and specific for the 9 steroid hormones analysis in human plasma and urine.关键词
