摘要

Abstract

Objective To study the in vivo effects of ligustrazine(TMP) on blood-brain barrier (BBB) and expression of Claudin-5 in rats after brain ischemia-reperfusion (I/R).Methods The focal cerebral I/R rat model of middle cerebral artery occlusion (MCAO) was established by intraluminal suture.The change of the permeability of BBB was detected using Evans blue assay.The expression of Claudin-5 was determined by immunohischemistry assay,RT-PCR and Western blot.Results The BBB permeability was found significantly decreased at 3 h and 72 h after I/R compared with sham-operated group.The BBB permeability in ligustrazine group was found signiticantly increased compared with the group of I/R at the same time points.The expression of Claudin-5 was found significantly decreased at 3 h and 72 h after I/R group compared with sham-operated group.The expression of Claudin-5 in ligustrazine group was found significantly increased compared with the I/R group at the same testing time points.Conclusions The down-regulation of Claudin-5 is closely related to the BBB injury induced by cerebral ischemia-reperfusion.Ligustrazine contributes to the cerebral edema after reperfusion through the up-regulation of Claudin-5,recovery of function of tight junction and reducing the permeability of BBB.
