Fermentation optimization of methanol-tolerant lipase-producing strain Galactomyses geotrichum SXL-107

Abstract
In order to enhance the productivity of SXL-107 lipase,the culture condition of Galactomyses geotrichum SXL-107 stain was optimized.The single-factor experiment was used to screen carbon source,nitrogen and inducer; Response surface methodology was used to determine fermentation culture conditions,and verified tests were carried out by 250 mL shake flask culture and 10 L fermentor fermentation.The optimum culture medium were contained with soybean flour 25.56 g · L-1,olive oil 16.74 g· L-1,sucrose 4.78 g· L-1,peptone 10.00 g · L-1,(NH4)2SO4 5.00 g · L-1,KH2PO41.50 g · L-1and Mg2SO4,1.00 g · L-1.The fermentation temperature was 30 ℃ by initial pH6.0.The olive oil was added by three times (0,6,12 h) in the period of fermentation cultivation.Under the optimal conditions,the lipase activity improved from 104 U · mL-1 to 317.33 U · mL-1,enhanced by 3.05 folds under 250 mL shake flask culture.And the highest lipase activity of 332.03 U ·mL-1 was achieved by batch fermentation in a 10 L fermentor after 38 h of cultivation.关键词
