UPLC法测定人血清中去甲万古霉素的浓度
Determination of Norvancomycin in Human Serum by UPLC
摘要
Abstract
OBJECTIVE:To establish a method for the determination of norvancomycin concentration in human serum.METHODS:After treated with acetonitrile-6％ perchloric acid (1∶5),UPLC method was adopted to determine the concentration of norvancomycin in blood sample.BEH C18 column was used with mobile phase consisted of acetonitrile-0.05 mol/L potassium dihydrogen phosphate buffer (pH3.2,gradient elution) at the flow rate of 0.25 ml/min.The detection wavelength and column temperature were set at 236 nm and 35 ℃,respectively.Vancomycin was used as the internal standard.RESULTS:The linear range of norvancomycin was 2-100 μg/ml with recovery of 98.06％-99.82％.RSDs of inter-day and intra-day were all lower than 2％.CONCLUSIONS:The method is convenient,sensitive,accurate and suitable for the serum concentration monitoring of norvancomycin in the clinic.关键词
