Effect of FK506 on NIT-1 cells proliferation and insulin secretion
目的 探讨他克莫司(FK506)对NOD鼠胰岛β细胞系(NIT-1)增殖、凋亡和胰岛素分泌的影响.方法 用MTT法、流式细胞术检测NIT-1细胞增殖;放射免疫法、real-time PCR检测NIT-1细胞胰岛素分泌和胰岛素分化相关基因PDX-1、GLUT2和GK mRNA的表达.结果 FK506(20 μg/L)抑制NIT-1细胞增殖(P<0.05),诱导细胞凋亡(P<0.05);FK506(10 μg/L)抑制NIT-1细胞释放胰岛素(P<0…查看全部>>
Objective To investigate the effects of FK506 on proliferation of pancreatic NIT-1 beta-cell and insulin secretion.Methods Effect of FK506 on proliferation and apoptotic was mensured by MTT-assay,flowcytometry; insulin secretion was investigated by radioimmunoassay and real-time PCR.Results FK506 (20 ag/L) may inhibit the proliferation of NIT-1 cells,and induce apoptosis; FK506(10 μg/L) may inhibit NIT-1 cell insulin release,The mRNA expression of PDX-…查看全部>>
