急性染镉对大鼠心肌细胞闰盘超微结构和连接蛋白43表达的影响及黄芪甲苷的拮抗作用

摘要
Abstract
Objective:To observe the effect of cadmium (Cd) on cardiomyocyte intercalated disc ultrastructure and expression of connexin 43 (Cx43) in rats,and the protection mechanism of astragaloside Ⅳ (As Ⅳ) on it.Methods:SD rats were randomly divided into 3 groups:control group,Cd group (rats were treated with Cd 1 mg · d-1 · kg-1 body weight by intraperitoneal injection),Cd+As Ⅳ group (the dose of cadmium was 1 mg · d-1 · kg-1 body weight,the dose of As Ⅳ respectively were 5 mg· d-1 · kg.-1 body weight and 10 mg · d-1 · kg-1 body weight).The rats after 1-week or 2-week treatment were anesthetized for thoracotomy,the ventricular tissues were respectively adopted for light and electron microscope observation and irnrnunohistochemistry.Results:The expression of Cx43 was significantly reduced,and cardiac muscle fibers and intercalated disc ultrastructure were both damaged in Cd group; however,those in Cd+ As Ⅳ were improved when compared with Cd group.Conclusion:Cd can damage cardiomyocyte intercalated disc ultrastructure,and reduce expression of Cx43; astragaloside Ⅳ retains the expression level of Cx43 and maintains morphologic integrity of cardiomyocyte intercalated disc.关键词
