摘要

Abstract

Objective To investigate effect of Yizhi-Fangdai Decoction on the expression of Aβ 1-40-induced apoptosisrelated genes in rat hippocampus neural cells.Methods In this study,six animal groups were used,including blank control,model control,groups receiving different concentrations (mild,moderate and high) of herbs and Donepezil group.Experimental AD model was established by intraventricular injection of Aβ1-40.Total RNA of hippocampal neuron was extracted 4 weeks after treatment administration.Expression of Caspase-3,Bcl-2,Bax and p53 was evaluated using RT-PCR.Results Compared to blank control group,expression of Bcl-2 significantly declined,while Caspase-3,Bax and p53 significantly increased in experimental groups (P<0.01).Treatment with Yizhi-Fangdai decoction upregulated Bcl-2 level in hippocampal neural cells (P < 0.01),and down-regulated Caspase-3,Bax and p53 levels (P<0.01).Conclusions Elevated Bcl-2 expression and inhibited Caspase-3,Bax and p53 expression with Yizhi-Fangdai decoction administration could be a possible mechanism that Yizhi-Fangdai decoction improves learning and memory function.
