High-level arachidonic acid-rich oil accumulation in Mortierella alpina though three-stage fermentation strategy

Abstract
In order to improve the rate of arachidonic acid (ARA) production,a three-stage fermentation strategy was used by Mortierella alpina,based on the mechanism of ARA biosynthesis.In the first stage,cells were cultivated for propagation in the nutrient-rich medium.The highest cell accumulation rate was obtained when the initial glucose content was 60 g/L of the nutrient-rich medium.In the second stage,cells were cultivated to achieve high cellular lipid contents in the glucose-rich medium without other nutrients.The effects of glucose concentration,the inoculation age,pH and culture time on lipid production were studied and optimized.In the last stage,cells were cultivated to achieve high ARA contents.In this stage,ARA rapid accumulation was stimulated by the medium without glucose and other nutritions,and the highest ARA content was reached when the cultivation time was 36 h.After three-stage fermentation the dry cell weight,total fatty acid and ARA reached 41.6,26.6 and 11.4 g/L,respectively.The result compared with the batch fermentation had improved significantly.This was the first report of using a three-stage fermentation strategy in ARA production,and it could be applied to the other similar oleaginous microorganisms to achieve high related PUFAs accumulation.关键词
