摘要

Abstract

Objective To apply UV spectrophotometer for the diagnosis of paraquat poisoning and develop a method for qualitative and quantitative detection of serum paraquat by UV spectrophotometer.Methods The serum had its protein precipitated with 20% TCA,and then went through 10-min storage in the-20 ℃ refrigerator and 12 000 r/min high-speed centrifugation.The superuatant was measured by UV spectrophotometer,with 50 μL microcell used.Results Linear correlation occurred when the serum paraquat concentration was limited between 0.10 μg/mL and 40 μg/mL,with the regression equation of y=0.081 2x+0.0104,the coefficient correlation of 0.999 6,the recovery rate between 92.5% and 103.0%,relative standard deviation between(RSD) 3.6% and 4.7%.The RSD was between 2.8% and 4.8% in a day and between 2.2% and 4.6% in several days,with minimum detection limit of 0.05 μg/mL.Conclusion A simple and accurate method is provided for the qualitative and quantitative detection of serum paraquat of the poisoning patient,which can be used for paraquat concentration measuring before and after hemoperfusion.[Chinese Medical Equipment Journal,
