Abstract
Objective To study the protective effect and mechanisms of exogenous hydrogen sulfide (H2S,NaHS) on global cerebral ischemia reperfusion injury in rats.Methods A total of 18 healthy male SD rats were randomly allocated into sham operation,isotonic saline,and NaHS intervention groups (n =6 in each group).A rat global cerebral ischemia-reperfusion model was induced by using the modified four-vessel occlusion method.After ischemia-reperfusion in rats,the rats in the NaHS intervention group were intraperitoneally injected NaHS (25 μmol/kg) immediately,and the rats in the isotonic saline group were intraperitoneally injected isotonic saline,both once a day for 7 days.The rats in the sham operation group only expose the vessels and did not perform any other treatments.The levels of superoxide dismutase (SOD) and malondialdehyde (MDA) in rat hippocampal tissue were detected,and thionine staining was used to observe the neuronal density of hippocampal CA1 regions.Results ①The SOD levels of the isotonic saline group (133 ± 12 kU/g) and NaHS intervention group (229 ± 21 kU/g) were lower than that (297 ±24 kU/g) of the sham operation group,and the SOD level of the isotonic saline group was lower than that in the NaHS intervention group.The differences were statistically significant (P < 0.01).②The MDA levels of the isotonic saline group (55.4 ± 6.2 μmol/g) and the NaHS intervention group (22.5 ±7.9 μmol/g) were higher than that of the sham operation group (6.1 ± 1.3 μmol/g),and the MDA level of the isotonic saline group was higher than that of the NaHS intervention group.The differences were statistically significant (P < 0.01).③The neuronal densities of the isotonic saline group (88 ± 15/mm2) and the NaHS intervention group (156 ± 19/mm2) were lower than that of the sham operation group (237 ±25/mm2),and the neuronal density of the isotonic saline group was lower than that of the NaHS intervention group.The differences were statistically significant (P < 0.01).Conclusion H2S has protective effect for cerebral ischemia-reperfusion injury in rats.Its mechanisms may be associated with the increased antioxidase activities and anti-lipid peroxidation injury.关键词
