GPC3特异的siRNA筛选和对肝癌细胞增殖能力的影响
GPC3-specific siRNA screening and effect on proliferation of hepatocellular carcinoma cells
摘要
Abstract
Objective To screen GPC3 specific siRNA and study the mechanism of GPC3 function in the development of HCC. Methods siRNAs targeting GPC3 were disigned and transfected into huh7 cells,and the inhibiting effect in the cells was assayed by using real-time PCR. Then the siRNA with obvious inhibition effect on GPC3 expression was transfected into Huh7 cells,GPC3 protein expression level was detect by Western Blot and proliferation of huh7 cells was assayed by Edu method. Results We successfully selected an GPC3 specific siRNA with 74.87% inhibiting efficiency,which could significantly inhibit the transcription and translation of GPC3. Furthermore,we found that knocking down of GPC3 could inhibit the proliferation of huh7 cells. Conclusion GPC3 expression may enhance development of HCC,GPC3 specific siRNA may be used as a tool for GPC3 research.关键词
