摘要

Abstract

The correlation between endogenous IAA level and leaf phenotype was investigated by introducing tryptophan monooxidase gene (iaaM),or co-expression of iaaM and isopentenyl transferase (ipt) into tobacco (Nicotiana tabacum L.) plants via A.tumefaciens transfection.Semiquantitative RT-PCR,ELISA and microscopy were used,respectively,to analyze transgene expression level,to quantitate endogenous phytohormones (zeatin riboside-ZR,isopentenyl adenosine-IPA,indole-3-acetic acid-IAA),leaf stoma density,guard cell length,palisade tissue cell size and leaf cross-section.Data showed that in comparison with the wild type plants,transgenic plants-iaaM10 displayed the highest content in IAA but the lowest content in cytokinin; whereas iaaMipt3 showed the highest level in cytokinin.With increasing IAA levels,stomatal density decreased,whereas guard cell length,palisade tissue cell length and width,and thickness of leaf cross-section all increased.A possible effect of cytokinin-to-auxin ratio on plant phenotype was discussed.
