Abstract

In order to increase the proportion of mature schizonts and improve the efficiency of rodent malaria parasite transfection,the in vitro culture conditions of the Plosmodium berghei ANKA1596cl1 (Pb1596) were optimized in many aspects,for example,the state of the parasites when we collected,the culture environment and the period of experiment,and so on,all of which based on the observation of the characteristics of Pb1596 in a growth cycle.In the experiment,trophozoites infected erythrocytes were collected by density-gradient centrifugation when parasitemia is in the range of 5%-15%,and then they were cultured in small volume of complete medium (0.01 L) for 8-10 hours.Finally,proportion of mature schizonts was counted and compared with the traditional method,and the growth state of parasites was observed.The data shows that the in vitro culture conditions of Pb1596 were optimized,which resulted in a remarkably increasing of mature schizont proportion from 0.97% to 24.37% compared with the traditional method,and also increased the average number of merozoites in a schizont from 13 to 16,shorten the experimental period to 11-13 hours,and avoided the loss of free merozoites.At the same time,a high survival rate of 90.27% of the blood cells was guaranteed.So in brief,we have improved the in vitro culture conditions of Pb1596,and provide the possibility to increase the efficiency of the rodent malaria parasite transfection for the further research.
