Abstract

Objective To investigate the protective effect of pretreatment with erythropoietin (EPO) on renal injury following cardiopulmonary bypass (CPB),and to explore related mechanism.Methods Thirty male Sprague-Dawley rats were randomly divided into three groups:sham,CPB,and EPO (CPB + 3 000 U/kg EPO) groups.The flow-rate was maintained at ≥100 mL/(kg · min)for 1 h.Blood samples were collected at the beginning (T0) and the end of CPB (T1),and 0.5 (T2),1 h(T3),2 h (T4)and 24 h(T5) after CPB for assays of Cr and BUN.Renal samples were obtained 24 h after the operation for the determination of TNF-a,IL-1β,IL-6,MDA,CAT,SOD,and for histopathologic examination.Results Compared with those of the CPB group,the EPO group showed an obvious decrease in plasma of Cr and BUN except T0 (P < 0.05).Levels of TNF-a,IL-1β,IL-6 and MDA in renal tissue of the EPO treated animals were also significantly lower than those of the CPB group (P < 0.05).But the activities of SOD and CAT remained a marked increase in CPB group (P < 0.05).Renal impairment with cast formation and tubular necrosis in the tubular epithelium was found in CPB group.These changes were markedly reversed in EPO group.Conclusions EPO could have potential effect on preventing CPB-induced renal injury,probably through its anti-in ammatory effect and antioxidant function.
