Abstract

Objective To investigate the effect of hypothermia induced by adenosine 5'-monophosphate (5'-AMP) on endotoxemic rats.Methods Sixty male Wister rats were evenly randomized to four groups as control,lipopolysaccharide (LPS),5'-AMP-hypothermia therapy (HT),and 5'-AMP-hypothermia prevention (HP).An endotoxemia model was created by intraperitoneal injection of LPS in LPS group; For rats in HT group,5'-AMP was given for hypothermia intervention when the rat temper ature rose by 0.5 ℃ after intraperitoneal LPS; For HP group,a hypothermic animal model was created by injecting 5'-AMP followed by LPS for inflammation induction.ELISA method was applied to detect serum levels of inflammatory cytokine in rats at 3,6 and 12 hours after injection of LPS,their lung tissues were analyzed pathologically with hematoxylin-eosin staining.Results Compared with the rats in LPS group,the hypothermia induced by 5'-AMP could markedly decrease serum levels of TNF-α,IL-1β and IL-6 and increase IL-10 (F =2.50-26.73 ; q =6.36-214.85 ; P<0.05).Pathologically,hypothermia induced by 5'-AMP obviously inhibited acute lung injury induced by LPS.Conclusion Hypothermia induced by 5'-AMP plays a noticeable role in the prevention and treatment of endotoxemia and acute pulmonary injury in rats caused by LPS.
