Content Determination of Argatroban Injection by RP-HPLC
摘要
Abstract
OBJECTIVE:To establish the method for the content determination of Argatroban injection.METHODS:RP-HPLC was adopted.The determination was performed on Venusil ASB C18 column with mobile phase composed of methanol-0.002 5％ glacial acetic acid water solution (pH 5.0) (50:50) at the flow rate of 0.8 ml/min.The column temperature was set at 45 ℃ and the detection wavelength was 259 nm.The content of argatroban was determined by peak area of argatroban 21 R/S isomers.RESULTS:The retention time of argatroban 21 R/S isomers were 38.01 min and 40.49 min respectively.The linear range of argatroban was 39.84-149.40 μg/ml (r=0.999 8) with an average recovery of 99.7％ (RSD=0.64％).CONCLUSIONS:The method is simple,accurate,reliable and specific which can be used for the content determination of Argatroban injection.关键词
