摘要

Abstract

Objective: To explore the metabolite profile of mesenchymal stem cells (MSCs) underwent osteogenic differentiation using 9.4 T high resolution MR spectroscopy. Materials and Methods: Human umbilical cord mesenchymal stem cells were cultured and collected. MSCs were induced osteogenic differentiation for 2 weeks and cell metabolite extractions were prepared by methanol-chloroform (M/C) method. Cell extractions were then analyzed on a 9.4 T MRS device. Results:After osteogenic inducement, the fatty acid metabolite peak at 2.02 ppm and 2.25 ppm obviously decreased, and there appeared a new metabolite peak at about 1.77 ppm, which is most probably representative of leucine. Conclusions:There are some speciifc characteristics on MRS of MSCs underwent osteogenic differentiation. The new arised peak at 1.77 ppm may serve as the biomarker for mesenchymal stem cells osteogenic differentiation.
