番鸭细小病毒 ZQ 分离株非结构蛋白基因的克隆及序列分析
Cloning and Sequencing of NS Genes of Muscovy Duck Parvovirus ZQ Strain
摘要
Abstract
In this study,a pair of specific primers were designed based on the sequence of Muscovy duck parvovirus (MDPV)genes published in GenBank to amplifiy the NS1 and NS2 genes of MDPV ZQ stain by polymerase chain reaction (PCR).The fragment was linked to the vector pMD18-T and transformed in-tothe competent Escherichia coli DH5α.After enzyme digestion and polymerase chain reaction(PCR)u-sing plasmid as template,a recombinant plasmid was screened which consist of 1.9 kb/1.3 kb gene frag-ment.The result showed that the purpose fragmen of recombinant plasmid of NS1 consisted of 1 884 nu-cleotides and encoded 627 amino acids,and NS2 gene consisted of 1 356 nucleotides and encoded 451 amino acids.The NS1 gene shared 98.1%-99.6% homology with that of other isolates published at nueleotide level,and the NS2 gene homology was 98.2%-100%.关键词
