HAp/mSiO2载药颗粒的制备及体外药物释放行为

Abstract
Nano-sized spherical hydroxyapatite (HAp)particles were prepared via a wet chemical precipitation method after the solutions comprising calcium and phosphate precursors were mixed at a slow addition rate.According to the high adsorption ca-pacity for drug (methylene blue,MB)and the standard deviation,the optimal experimental condition of sol-gel route was deter-mined via the orthogonal experiment design method.The HAp/mesoporous silica (HAp/mSiO2 )powder was fabricated through the deposition of mSiO2 on HAp at low silanophilic activity under the optimal experimental condition.The adsorption capacity of MB onto HAp/mSiO2 in solutions containing MB at a concentration of 0.10 g/L is 79.26 mg/g.The adsorption iso-therm of methylene blue onto HAp/mSiO2 is a type II isotherm,i.e.,the monomer and dimmers of drug molecules are ad-sorbed on HAp/mSiO2 .The sustained-release behavior of drugs from HAp/mSiO2 is related to the pH value of the buffers (pH=7.2~7.4 in phosphate buffered saline and pH=4.7 in lysosome-like buffers)due to the influence of solution properties on the microstructures of drug carriers and interaction mechanisms between drug molecules and powders.关键词
