Abstract

Objective:To observe the effect of single intraviteal injection of CNTF on the axonal regeneration of retinal ganglion cells in the autologous sciatic nerve transplants in golden hamsters. Methods: Twenty golden hamsters were randomly divided into normal control group (n=2), control group (A group, n=6), CNTF 2 μg group (B group, n=6) and CNTF 1μg (C group, n=6). In all the hamsters, a segment of autologous sciatic nerve was transplanted to the orbital end of the transected optic nerve. The animals in the groups A, B, and C were immediately intravitreally injected with 2 μL of normal saline, 2 μL CNTF solution containing 2μg CNTF, and 2μL CNTF solution containing 1μg CNTF respectively. The regenerating retinal ganglion cells (RGCs) was visualized by flouro-gold and the labeled RGCs were counted. Results: The number of regeneration of retinal ganglion cells of the groups A, B, and C were 4932, 8596, and 7338 respectively. The number of labeled ganglion cells in the groups B and C were significantly higher than that of the group A ( <0.05). There was no statistical difference between B and C groups. Conclusion:Single intravitreal injection of CNTF can markedly increase the axonal regrowth of RGCs in sciatic nerve transplants in hamsters.
