Effect of HERG Channel Blocker on Proliferation of Gastric Cancer Cells

Abstract
Objective To investigate the effect of HERG channel blocker on the proliferation of gastric cancer cells.Methods HERG channel blocker was used to interfere HERG current of gastric cancer cells ,and then the growth curve of gastric cancer cells was drawn by MTT method;The ability of clone formation of gastric cancer cells was studied by clone formation assay;The cell cycle distribution of gastric cancer cells was investigated by flow cytometry .Results The proliferation of gastric cancer cells was inhibited in a time-and dose-dependent manner when gastric cancer cells were treated with HERG channel blocker ( cisapride ) ,but cisapride had little effect on proliferation of gastric epithelial cells.The clonogenicity of gastric cancer cells treated with cisapride decreased compared with control group (P<0.05). Cisapride tended to inhibit gastric cancer cells entering S phase from G 1 phase in the cell cycle .Conclusion HERG channel blocker can inhibit the proliferation , clone formation and progression of G 1/S of gastric cancer cells , which indicates that HERG current may play a role in the proliferation of gastric cancer cells ,and HERG protein is a potential target for gastric cancer biological therapy .关键词
