Study on Effects of Osteopontin on Post-Transcriptional Regulation of MMP-9 Gene Expression in PANC-1 Cells

Abstract
Objective To investigate the effects of osteopontin on post-transcriptional regulation of matrix metalloproteinase-9(MMP-9) gene expression in PANC-1 cells.Methods Real-time PCR and luciferase reporter gene assay were used to detect the effects of osteopontin on post-transcriptional regulation of MMP-9 gene expression in PANC-1 cells.Results Osteopontin prolonged the half-life of MMP-9 mRNA and up-regulated the expression of luciferase in report gene plasmid which contained 3′UTR of MMP-9 mRNA in PANC-1 cells.Conclusion Osteopontin activates the expression of MMP-9 partly through affecting post-transcriptional regulation of MMP-9 gene expression in PANC-1 cells, whose primary target is 3′UTR of MMP-9.关键词
