Effects of HERG Protein on Cell Cycle and Apoptosis of Gastric Cancer Cells

Abstract
Objective To explore the effects of human ether-a-go-go-related gene ( HERG ) protein on cell cycle and apoptosis of gastric cancer cells .Methods HERG-siRNA vector was constructed and transfected into gastric cancer cell SGC7901 ,G418 was applied to screening and cloning the transfected cells which were verified with HERG current and protein .Flow cytometry was used to assess the apoptosis of gastric cancer cells;Transmission electron microscope was used to observe the ultrastructure of gastric cancer cells ;The cell cycle distribution of gastric cancer cells was investigated by flow cytometry .Results HERG-siRNA eliminated the HERG current in gastric cancer cells and induced apoptosis in gastric cancer cells;There were typical changes of apoptosis under transmission electron microscope;HERG-siRNA inhibited gastric cancer cells entering into S phage from G 1 phage.Conclusion HERG-siRNA can inhibit HERG protein and potassium current as well as the progression of G 1/S of gastric cancer cells ,and induce apoptosis of gastric cancer cells .HERG -siRNA has potential anti-cancer effects on gastric cancer cells .关键词
