麻疯树亚硝基谷胱甘肽还原酶(JcGSNOR)的克隆与功能分析
Molecular clone and functional analysis of S-nitrosoglutathione reductase/GSNOR from Jatropha curcas

Abstract
JcGSNOR was cloned by RT-PCR and RACE techniques from Jatropha curcas .The expres-sion situation of JcGSNOR in different organs of Jatropha curcas was detected by real time PCR .The result showed that ,JcGSNOR was expressed in all tissue detected .The expression level was the highest in the seed ,root and stem were more or less ,and leaf was the least .With bioinformatic analysis ,the JcGSNOR may be comprised of 379 amino acids ,to give the molecular weight of 40 .256 kDa and the pI value of 6 .74;Result analyzed by NCBI showed that ,JcGSNOR has up to 80% similarity with GSNOR of other plants ,which indicated JcGSNOR is a new member of GSNOR family .To identify the biochemi-cal activity of JcGSNOR ,the gene encoding JcGSNOR was cloned into saccharomyces cerevisiae .GSN-OR activity and growth curve of yeast were detected .This study indicated that ,JcGSNOR showed no obvious effect on grow th of yeast ,but enhanced the GSNOR activity significantly .关键词
