环氧合酶-2基因敲低对足细胞相关蛋白表达的影响
The effect of COX-2 gene knockdown on podocyte associated proteins
摘要
Abstract
Objective:To investigate the effect of cyclooxygenase-2 (COX-2) gene knockdown on podocyte associated proteins .Methods:The conditionally immortalized murine podocytes were divided into three groups :nor-mal control group ,podocytes treated with siRNA and podocytes treated with negative siRNA .The level of podocyte apoptosis was measured by flow cytometry .The proteins expression of COX-2、Nephrin、Podocin、CD2AP were detec-ted by western blotting .Results :The expression of COX-2 protein was decreased and the podocytes apoptosis were increased in podocytes treated with siRNA .And the protein expressions of Nephrin、Podocin、CD2AP in siRNA group were significantly lower than those in normal control group and negative siRNA group .Conclusion :The COX-2 gene knockdown induces podocytes apoptosis .The protein expression of Nephrin、Podocin、CD2AP in podocyte treated with siRNA are downregulated .关键词
