Abstract

Objective To investigate the effects of glypican-3 (GPC-3)gene knockdown by specific short hairpin RNA(shRNA)on proliferation and apoptosis of HepG2 cells. Methods Four pairs of GPC-3 shRNA were designed,synthesized and screened according to the inhibiting efficiency and plasmids containing specific GPC-3 shRNA were constructed;HepG2 cells were transfected with constructed plasmid containing GPC-3 shRNA with the highest inhibiting rate,and cell proliferation,cell cycle transition,and apoptosis were determined after GPC-3 knockdown. Results Four plasmids containing specific GPC-3 shRNA were successfully constructed,and one of which had the highest inhibiting rate by reaching to 89.3%;The proliferation rates of the HepG2 cells transfected with the efficient GPC-3 shRNA decreased to 33.2%,56.0% and 71.1%,respectively,as compared to the corre-sponding controls(P<0.05) 24h,48h and 72 hours after transfection;The cell cycles were arrested at G1 phase af-ter GPC-3 knockdown,while the apoptosis rate of transfected HepG2 cells increased to 65.6%,significantly higher than that in control (5%,P<0.05). Conclusions Specific GPC-3 shRNA can effectively knockdown GPC-3 gene transcription in HepG2 cells,resulting in cell proliferation inhibition,cell cycle arrest and increased apoptosis;GPC-3 might be a potential target gene for hepatocellular carcinoma therapy.
