Abstract

Benzo(a)pyrene (BaP), with five benzo-rings,is an important isomers of polycyclic aromatic hydrocarbons (PAHs), and it is a prevalent pollution substances in environment. The mechanism of environmental toxicology of BaP is DNA damage. Comet assay, named Single Cell Gel Electrophoresis (SCGE) is a sensitive, fast technology to detect DNA ring damage;this technology is broadly applied to bio-monitor in aquatic environment. The toxicological monitoring and assessment of pollutant is always a hotspot of environmental research. This eco-toxicological experiment focused on the DNA damage of tilapia (GIFT Oreochromis niloticus) when exposed to BaP via comet assay.Tilapia (GIFT Oreochromis niloticus) were exposed to different concentrations of ben-zo(a)pyrene (0.1 µg·L-1 ,1 µg·L-1 ,0 µg·L-1 , 50 µg·L-1 ) for 4 d, 7 d and 14 d respectively. DNA damage in liver cells of tilapia was evaluated by comet assay. Tail DNA content, tail length and tail moment, Olive tail moment were used to evaluate DNA damage. Results showed that BaP exposure could cause liver cell DNA damage in tilapia. Among the concentration of 0.1 µg·L-1, 1 µg·L-1, 10 µg·L-1, DNA damage increased following the BaP exposed concentrations, but decreased in 50μg·L-1 group. In addition to the tail DNA content of 1 µg·L-1 dose group, tail DNA and Olive tail moment of 10μg·L-1 group, other parameters decreased in 7 d and then increased. These results demonstrated that liver cell DNA damage can reflect BaP pollution of water environment to a certain extent. The experiment provided a detailed data to understand the mechnism and environmental monitoring of BaP.
