Smad3磷酸化促进溃疡性结肠炎的黏膜修复
Smad3 phosphorylation contributes to the recovery of ulcerative colitis

Abstract
Objective To investigate the process of mucosal injury and repair of ulcerative colitis in mice and the role of p-Smad3 which is the key factor in transforming growth factor-β(TGF-β)signaling path-way.Methods Mice were exposed to 3%dextran sulfate sodium (DSS)for 5 days to induce acute ulcerative colitis,then followed with water for 4 weeks for mucosal repair.Mice were divided into six groups:control group that were allowed to drink only water,injury group that were exposed to 3%DSS for 5 days and the other four recovery groups which drunk water for 1 ,2,3,4 weeks after DSS removal,respectively.General health condition was recorded daily and histological examination was performed to analysis clinical and histopathologi-cal changes of DSS-induced colitis.Related markers of apoptosis and proliferation were detected.The expres-sion of TGF-βand p-Smad3 were measured to observe the change of TGF-βsignaling pathway during the muco-sal injury and repair process.Results Disease activity index of mice increased after DSS treatment and re-turned to normal at 2 weeks after DSS removal.The histological score significantly increased in injury group and began to decrease after 2 weeks of recovery.The apoptotic index rose to the maximal level in injury group and back to normal at 3 weeks after DSS withdrawal.However,the proliferation index reduced to the minimal level in injury group,then started to increase to the peak at 3 weeks.TGF-βexpression increased in all experimental groups,while the activation of Smad3 was inhibited in injury group,then began to be reactivated at 1 week af-ter DSS removal.Conclusion Smad3 phosphorylation promotes the repair of colonic mucosa of ulcerative coli-tis in mice.关键词
