绿色荧光蛋白标记的鼻咽癌放射抗拒性裸鼠移植瘤模型的建立及其活体成像观察
Establishment and dynamic in vivo imaging of a radioresistant,GFP-labelled xenograft tumor model of human nasopharyngeal carcinoma in mouse

Abstract
Objective To establish a GFP-labelled radioresistant xenograft tumor model of human nasopharyngeal carcinoma(NPC) and build the foundation for further study. Methods Lentivirus was used to establish a stable,radioresistant human NPC cell line expressing GFP(GFP-CNE-2R)and a corresponding non-radioresistant cell line(GFP-CNE-2).Both lines were subcutaneously implanted into BALB/c-nu nude mice;when the tumors had reached 1 cm in size,the animals were irradiated with 10 Gy in 1 fraction.Tumor proliferation and metastasis were analyzed in vivo using dynamic imaging technology. Results GFP-CNE-2R and GFP-CNE-2 lines were successfully established,and clone formation assays showed GFP-CNE-2R cells to be radioresistant.Xenograft tumors were grown from GFP-CNE-2R and GFP-CNE-2 cells,and growth of GFP-CNE-2R tumors was not significantly inhibited by radiation. Conclusion GFP-CNE-2R xenograft tumors are more radioresistant than GFP-CNE-2 xenograft tumors.关键词
