Abstract

Objective To investigate the effect of UA on the apoptosis and expressions of Bcl-2/bax in HL-60 cells. Methods HL-60 cell line was cultured in vitro. MTT methods was used to detect the inhibitory effect of UA on proliferation. Flow cyetmetry was used to observe the effect of UA on the apoptosis and cycle distribution of HL-60 cells. The influence of UA on the bcl-2 and bax protein expression of HL-60 cells was analyzed with western blot. Results UA markedly inhibited proliferation of HL-60 cells in a concentration-dependent manner, UA induced apoptosis of HL-60 cells in a concentration dependent manner,and decreased the proportion of cells in the G1 phase. UA can efficiently up-regulated bax and down-regulated bcl-2 protein expression of HL-60 cells in a concentration dependent manner. Conclusion UA inhibited proliferation and induced apoptosis on HL-60 cells partly by up-regulating expression of bax and down-regulating expression of bcl-2.
