Abstract
Objective To prepare bacitracin liposom,and to determine bacitracin content and entrapment efficiency of the liposome using HPLC method. Methods The bacitracin liposome was prepared by film dispersion method; The chromatographic conditions were Column: Diamonsil-C18 column(150mm×4.6mm,5μm); mobile phase: pH 6.0 phosphate buffer: methanol (60∶40); column temperature: 25℃; detection wavelength: 254nm; flow rate: 1mL/min.Refrigerated centrifugation using bacitracin liposomes free drug. Results The mean encapsulation efficiency of the bacitraci liposome was 87.94%(RSD 1.12%,n=5);In the chromatographic conditions bacitracin and accessories and a good separation of the solvent peak,bacitracin at 0.061-1.83μg range had a good linear relationship (R2=0.9997),recovery was 99.68%-101.38%. Conclusion The HPLC method is simple, accurate and reliable,can be used to determine bacitracin content and entrapment efficiency of the bacitracin liposom.关键词
