Abstract

Vibrio alginolyticus is one of the important pathogens causing Vibriosis in the marine aquaculture in South China. The acfA gene is the important virulence gene, and the expressed product is the necessary protein which is required for efficient intestinal colonization. Based on the sequences of the highly conserved DNA area of the acfA gene among vibrios, primers for PCR were designed. The partial sequence of the gene was amplified by touchdown PCR, then the flanking sequence was amplified by inverse PCR and nested PCR. The complete cDNA sequence of acfA gene was 743 bp and the complete ORF length was 648 bp which encoded a protein with 215 amino acid residues. A classic-35 region, -10 region and Shine-Dalgarno sequence couldn’t be recognized in upstream in the acfA gene, and the 18aa signal peptide could be found in the deduced amino acid sequence of the acfA gene. The analysis of protein structure showed ACFA protein mainly constructed byα-helix and random coil. Blast analysis indicated the deduced amino acid sequence of ACFA from V. alginolyticus showing high homology with Vibrio.
