Abstract

Objective To investigate the differences of functional connectivity on resting- state functional magnetic reso-nance imaging (fMRI) in elderly subjects with different cognitive function. Methods Thirteen patients with Alzheimer's disease (AD), 12 patients with mild cognition impairment (MCI) and 13 healthy elderly subjects (controls)underwent resting- state fMRI examinations with a 3.0 T MR system. Regional homogeneity (ReHo) was used to define the seeding voxel and to analyze the imaging data with resting- state functional connectivity. Results Increased connectivity of ReHo was detected in posterior cin-gulated cortex (PCC), medial prefrontal cortex (MPFC), dorsolateral frontal cortex and parietal cortex in AD and MCI patients. Compared to the controls, patients with MCI showed significantly reduced connectivity of ReHo in left temporal lobe (inferior and middle temporal gyrus), parahippocampal gyms and lingual gyms; while increased connectivity of ReHo was shown in right frontal lobe (superior frontal gyrus, middle frontal gyrus and precentral gyrus), superior parietal lobule, and left fusiform gyrus, su-perior temporal gyrus, and inferior frontal gyrus. Altered functional connectivity was found more prominent in AD patients than that in MCI patients. Significantly reduced connectivity of ReHo was detected between prefrontal- parietal lobe, temporal- parietal lobe, and temporal- occipital lobe;while increased connectivity of ReHo was shown within prefrontal lobe, and between occipi-tal- temporal lobes, thalamus- temporal lobes. There were significant differences among these three groups. Conclusion Differ-ent functional connectivity of ReHo among the patients with AD, MCI and normal elderly people might be used to differentiate the three different cognitive conditions.
