Abstract

OBJECTIVE:To evaluate the quality ofAtractylodes macrocephalae from the type of chemical component and peak area of characteristic component.METHODS:The HPLC fingerprint of ethanol extract from A.macrocephalae was established by HPLC.The determination was performed on CAPCELL PAK C18 (250 mm×4.6 mm,5 μm)column with mobile phase consisted of water-methanol (gradient elution) at the flow rate of 1.0 ml/min.The column temperature was 35℃,and detection wavelength was set at 220 nm.The sample size was 20 μl.TCM fingerprint similarity evaluation system,peak overlap rate calculation and hierarchical cluster analysis were used to assess it completely.RESULTS:10 common fingerprint peaks were detected in ethanol extract from 10 batches of A.macrocephalae,and atractylenolide Ⅲ,atractylenolide Ⅰ and atractylone were identified.The fingerprint peak overlap rate of A.macrocephalae ethanol extract from different producing areas was over 96%,and the similarity was over 90%.CONCLUSIONS:The established method can be use to evaluate the similarity ofA.macrocephalae from different producing areas in terms of chemical components and characteristic peak intensity,and provides foundation for the quality evaluation of A.macrocephalae and other medicinal materials.
