Effect of deubiquitinase USP14 on the proliferation of rat cardiac fibroblasts

Abstract
Objective To evaluate the effect of inhibiting deubiquitinase USP14 activity on the proliferation of cultured rat cardiac fibroblasts (CFs).Methods Cultured CFs from neonatal SD rats were isolated by trypsinization and differential centrifugation,treated with various dose of IU1,the inhibitor of deubiquitinase USP14,and examined after 48 h.The proliferations of CFs were detected by MTS,cell counts and crystal violet staining,the CFs cellular toxicity of IU1 was determined by LDH release detection.Result IU1 could inhibit the proliferation of CFs in a concentration-dependent manner and had no significant cytotoxicity.Conclusion The inhibitory activity of deubiquitinase USP14 can suppress the proliferation of CFs,deubiquitinase USP 14 may be an important target of anti-cardiac fibrosis.关键词
