可溶性CD40配体对NB4细胞增殖及survivin mRNA表达的影响

Abstract
Objective To investigate the effect of soluble CD40 ligand (sCD40L) on proliferation of NB4 cells and expressions of interleukin 6 (IL-6) and survivin mRNA.Methods MTT assay was used to examine the effect of different concentrations of sCD40L on leukemia NB4 cells proliferation in vitro at different incubation times;RT-PCR was used to determine the effect of different concentrations of sCD40L on the expression of survivin mRNA in NB4 cells at different incubation times;ELISA was used to detect cytokine IL-6 level in the culture supematant of leukemia NB4 cells of different concentrations of sCD40L at different incubation times.Results MTT assay showed that sCD40L with different concentrations (0.5-2.5μg/ml) could significantly inhibit the proliferation of NB4 cells in a dose-and-time dependent manner (P＜0.05).sCD40L could suppress the expression of survivin and IL-6,and with dose-and-time dependent manner,with statistically significant difference compared with no-treatment control group (P＜0.05).Conclusions sCD40L can significantly inhibit the proliferation of NB4 cells in vitro,yet it is concentration and time dependent.The mechanisms of sCD40L in inhibiting the proliferation of NB4 cells and inducing apoptosis in NB4 cells may be related to their inhibition of IL-6 and survivin expression.关键词
