聚羟基脂肪酸酯和聚酰胺-胺树状高分子构筑的经皮给药系统
Transdermal drug delivery system constructed by polyhydroxyalkanoates and PAMAM dendrimers

Abstract
BACKGROUND:Polyhydroxyalkanoates (PHA) can be used as anin vitro drug delivery carrier for its characteristics of viscosity and ability of drug mixture and dispersion. PAMAM dendrimers have unique physical and chemical properties as a drug carrier. <br> OBJECTIVE:To mix PHA with PAMAM to build a transdermal drug delivery system for indissolvable drugs. <br> METHODS:Pseudomonas putida KT2442 was cultured in a culture medium containing NH4Cl at 30℃ for 72 hours, then 5 g/L octane acid was added in the culture system and cultured for 24 hours, and PHA compound was extracted with chloroform. PHA compound matrix (10 mg) and PHA compound matrix containing PAMAM and phenethylamine dendrimers (10 mg) were respectively mixed with 5 mg of sulfonamide derivative Tamsulosin to construct a transdermal delivery system. Meanwhile, anin vitro permeation device was established to study the promoting effect of compound matrices on drug penetration. X-ray diffraction analysis was used to analyze the atomic spatial distribution of Tamsulosin, PHA, PHA compound matrix-Tamsulosin, and PHA compound matrix containing PAMAM dendrimers-Tamsulosin. <br> RESULTS AND CONCLUSION: PHA and mixture of PHA and PAMAM dendrimers could promote the transdermal effect of indissolvable drugs as a transdermal drug matrix. In the same condition, the mixture of PHA and PAMAM dendrimers could satisfy the clinical blood drug concentration as a transdermal drug matrix. This mixture can make drug model crystalized wel in a highly ordered manner.关键词
