磁性Fe3 O4纳米颗粒在家兔体内的分布及其药代动力学研究
Tissue Distribution and Pharmacokinetics of Magnetic Nano-Fe3 O4 in Rabbits

Abstract
Objective To study ultramicrostructure tissue distribution, concentration-time curve and effects on serum biochemical parameters of magnetic nano-Fe3 O4 in rabbits. Methods Magnetic nano-Fe3 O4 was injected through ear vein of rabbit. The blood was got at different times after the intravenous injection. The concentration of Nano-Fe3 O4 in rabbit blood was tested by ICP-OES and the pharmacokinetic parameters were calculated. Another 24 rabbits were divided into four groups randomly according to body weight,normal control group,low-dose group, middle-dose group, high-dose group. Magnetic nano-Fe3 O4 of different doses was injected through ear vein. The blood and different organs were got at 24 hours after the intravenous injection. The distribution of Nano-Fe3 O4 in different organs were tested and ultramicrostructure liver and spleen tissues were observed by TEM, biochemical parameters of serum in rabbits were tested. Results The concentration of nano-Fe3 O4 in rabbit blood was decreased with the time,which was consistent with two compartment model of pharmacokinetic. The distribution of nano-Fe3 O4 in different organs showed its own characteristics. It was mainly distributed in spleen and liver,and its distribution was different according to different doses in the same organ. The measurement of serum biochemical parameter contents showed some changes in ALP,AST and URIC of blood. Conclusion Nano-Fe3O4 is mainly distributed in spleen and liver of rabbits and mainly concentrated in macrophage in ultramicrostructure. There is no too much change in biochemical parameters of serum.关键词
