摘要

Abstract

Objective:Observe the influence of cordyceps sinensis( CS) on MIN6 cell apoptosis induced by IL-1βand its relationship with reactive oxygen species and UCP-2 in order to investigate the protective effects and mechanism of islet cells. Methods:The experi-ment groups were divided into normal control group,IL-1 beta group,IL-1β+CS low concentration group and IL-1β+CS high concen-tration group. The cell activity was tested by CCK8. The basal insulin of MIN6 cells and acute insulin response irritated by glucose were tested by Method Elisa. The cell apoptosis was detected by Hochest dyeing and flow cytometry. The reactive oxygen species were detec-ted by chemical fluorescence method. The expression of uncoupling protein 2 was detected by Western Blot. Results:Compared with the normal group, the cell activity and insulin secretion of IL-1β group reduced significantly. The cell apoptosis increased significantly, Meanwhile,the ROS activities increased and the UCP-2 decreased. After the low and high concentration CS were given,compared with the damage group,the cell activity and insulin secretion increased obviously. The apoptosis rate reduced significantly. The UCP-2 con-tent increased and the ROS activity decreased at the same time. Conclusion:CS would increase the cell activity and insulin secretion. It reduces the MIN6 cell apoptosis induced by IL-1β. The mechanism may be related with the increasing of expression of UCP-2 and the reduction of ROS activity.
