抑制ANGPTL4基因对舌癌细胞放射敏感性的影响
Effect of inhibited ANGPTL4 on radiosensitivity in tongue carcinoma cell line
摘要
Abstract
Objective:To explore the effect and relative mechanism of ANGPTL4 inhibited by antisense technology on radiosen-sitivity in tongue carcinoma cells. Methods:Tongue carcinoma cell line Tca8113 was taken as a research object. There were four groups in the study:the antisense oligodeoxynucleotide( ASODN) group,the sense oligodeoxynucleotide( SODN) group,the lipofectamine group and the blank control group. After ANGPTL4 ASODN transfection,the level of the ANGPTL4 expression in tongue carcinoma cells in ev-ery group was evaluated by means of RT-PCR. After 6MV X-ray irradiation,the ellular response to irradiation in tongue carcinoma cells was detected by colony forming test,the apoptosis rate in respective group was observed by fluorescent staining,the expression of Bcl-2 protein in respective group was determinated by flow cytometry. The single factor analysis of variance was used in the statistical process-ing. Results:After the transfection of ANGPTL4 ASODN,the ANGPTL4 expression in the ASODN group was suppressed,went down a-bout 44% compared with the blank control group,while no significant difference among the other three groups. After X-ray irradiation, the colony forming efficiency in the NGPTL4 ASODN group was significantly lower than that in the SODN,the lipofectamine and the blank control group(F=38. 782,P<0. 001);the apoptosis rate increased significantly(F=156. 320,P<0. 001);the expression of Bcl-2 protein reduced significantly(F=302. 895,P<0. 001). Conclusion:ANGPTL4 ASODN sensitized the Tca8113 to ionizing irra-diation through decreasing the expression of ANGPTL4,which might be related to down-regulating the expression of Bcl-2 and increasing the apoptosis rate of Tca8113 cells.关键词
