喹乙醇在鲫鱼体内的药物代谢动力学研究
Study on Pharmacokinetics of Olaquindox in Carassius auratus
摘要
Abstract
The concentration of olaquindox in the plasma of Carassius auratus was determined by high per-formance liquid chromatography tandem mass spectrometry (LC-MS/MS).Healthy Carassius auratus, were randomly divided into 2 groups,20 tail each group of ,30 mg per kg body weight oral dose of olaquin-dox was used,the blood samples were collected.The plasma was treated with 30 mL/L trichloroacetic acid concentrations for precipitation and detection of protein,statistical software 3P97 was used for processing plasma concentration-time data.It's best to fit the olaquindox concentration-time data to one-compartment open model with first order absorption after single oral dosing.The pharmacokinetic parameters of ola-quindox in Carassiusauratus are as follows :the elimination half-life T1/2Ke is 5.97 h±1.06 h ,the distri-bution half-life T1/2ka is 1.09 h±0.14 h ,the peak time Tmax is 3.25 h±0.41 h ,the peak concentration CTmax is 25.73 mg/L±2.53 mg/L,and the area under the curve (AUC)is 325.35 mg/(L·h)-1 ±69.01 mg/(L·h)-1 .The results showed that olaquindox was rapidly and completely absorbed and slowly elimi-nated after oral administration to Carassius auratus.关键词
