摘要

Abstract

To understand the character of NOBOX genes of yak ,two pairs of primers for NOBOX genes of yak was designed according to the published NOBOX genes sequence of Bos taurus in GenBank ,then se‐quencing and splicing ,and succeeded in cloning NOBOX genes of yak .The results of sequence analysis show that the length of the cloning NOBOX cDNA is 1 975 bp ,containing 1 500 bp open reading frame (ORF) .It encodes a peptide of 499 amino acids whose molecular mass is 53 .12 ku .Using sequence homol‐ogy analysis ,NOBOX genes are more conservative ,the gene sequence of yak and Bos taurus have the high‐est homology which is 97 .0% respectively .On this basis ,the experiment uses Mega 5 .0 software with NJ algorithm to construct a phylogenetic tree NOBOX protein ,analyzed of the evolutionary relationships a‐mong different species .The successful cloning of NOBOX gene sequence of yak provide important basis for the study of the molecular mechanism of Maiwa Yak oocyte maturation and its early embryonic develop‐ment ,as well as the protection of genetic resources and breeding .
