牛布鲁菌侵染对 HPT-8细胞 p38基因转录水平的影响
Effects of Brucella abortus Infection on p38 Gene Transcription Levels in HPT-8 Cells
摘要
Abstract
This study explored the effects of Brucella infection on p38 gene expressions in human placenta trophoblastic cells(HPT-8).HPT-8 cells were infected with Brucella abortus 2308 strain and RB51 strain respectively,the cells were collected at different infected time(0,4,8,12,48 h),and the total RNA was iso-lated and then cDNA obtained.Cloning vectors of p38ɑ/p38β/GAPDH genes were constructed and their standard curves were drew,and transcription levels of p38ɑ/p38β genes were measured by RT-qPCR.Re-sults showed that cDNA of HPT-8 cells befor and after Brucella infection,cloning vectors of pMD18-T-p38 ɑ/p38 β/GAPDH and standard curves were obtained.RT-qPCR analysis showed that compared with 0 h,the relative expressions of p38 ɑand p38 βgenes in HPT-8 cells that infected with Brucella 2308 strain and RB51 strain with different times(4,8,12,48 h)were decreased markedly,and the difference was signif-icant(P <0.01).But the statistics results had some difference since they came from different strains.All of these results indicated that p38 MAPK signal pathway was down regulated in Brucella infected HPT-8 cells,meaning that p38 MAPK signaling pathways is involved in the regulation of biological effect caused by Brucella,and this regulation is related with the strength of the virulence of the strains,the greater viru-lence,the stronger regulation.关键词
