Effect of Ad-PTEN on the Proliferation and Apoptosis of 786-O Cells
摘要
Abstract
Objective:To observe the expression of PTEN in 786-O cells,which are mediated by recombinant adenovirus Ad-PTEN,and to investigate the inhibition of cell proliferation and migration,and induction of apoptosis by Ad-PTEN. Methods:The efficiency of adenovirus infecting 786-O cells was determined by observation of GFP expression.After the expression of PTEN protein was detected by westernblot,cell proliferation and migration was tested by MTT and cell dam-age healing experiment respectively.Cell apoptosis was observed by PI staining.Results:The efficiency of adenovirus in-fected 786-O cells for 48h was 100%,when the MOI was 100.After PTEN protein expression was confirmed by west-ernblot,cell proliferation was inhibited significantly (P<0.05 ),and cells were repressed to migrate from scratch site. And also Ad-PTEN induced the apoptosis.Conclusion:The recombinant adenovirus Ad-PTEN can mediate 786-O cells expression of PTEN protein efficiently,repress cell proliferation and migration,and induce apoptosis,which provides a theoretical basis for gene therapy on kidney cancer.关键词
